Can Simple DNS Plus be run as a Windows Service (a.k.a. "NT service")?

Yes Simple DNS Plus v. 5.0 and later always runs as a Windows service - this is not optional.

In earlier versions of Simple DNS Plus, this is an option which has to be enabled in the Options dialog / General tab.
